Overview

MOTS-c is a 16-amino-acid peptide encoded in mitochondrial DNA, first described in 2015 as a regulator of metabolism and insulin sensitivity.

Mechanism

In the original study its primary target was skeletal muscle, where it inhibited the folate cycle and activated AMPK, an energy-sensing enzyme.

    Discovery study (Cell Metabolism, 2015)

    In mice, MOTS-c treatment prevented age-related and high-fat-diet-induced insulin resistance and diet-induced obesity.[1]

    Human trials

    A phase 2 trial of MOTS-c for insulin sensitivity in prediabetes is recruiting (NCT07505745). An analog, CB4211, completed a phase 1a/1b study (NCT03998514).[2][3]
