Research summary

Semaglutide

Regulatory status
FDA-approved medicine: Ozempic (type 2 diabetes, Dec 2017) and Wegovy (chronic weight management, Jun 2021).
Strongest evidence
Approved drug backed by large phase 3 and outcome trials
Overview

Semaglutide is a long-acting GLP-1 receptor agonist made by Novo Nordisk, sold as Ozempic, Wegovy and the oral Rybelsus.

What researchers study it for
  • Type 2 diabetes
  • Obesity and overweight
  • Cardiovascular risk in people with obesity
Mechanism

Activates the GLP-1 receptor: glucose-dependent insulin release, slower gastric emptying and reduced appetite.

Evidence (3)
  1. Human · randomized trial

    STEP 1 (phase 3, NEJM 2021)

1,961 adults with obesity or overweight without diabetes, 68 weeks. Mean weight change −14.9% versus −2.4% with placebo; nausea and diarrhoea were the most common adverse events.[1]

  2. Human · randomized trial

    SELECT (cardiovascular outcomes, NEJM 2023)

17,604 people with cardiovascular disease and overweight or obesity but no diabetes. Major cardiovascular events occurred in 6.5% with semaglutide versus 8.0% with placebo (hazard ratio 0.80); stopping because of adverse events was more common with semaglutide (16.6% vs 8.2%).[2]

  3. Approved drug

    FDA approvals

    Ozempic approved Dec 5, 2017; Wegovy approved Jun 4, 2021 (Drugs@FDA).[3][4]
