Research summary

Tirzepatide(LY3298176)

Regulatory status
FDA-approved medicine: Mounjaro (type 2 diabetes, May 2022) and Zepbound (chronic weight management, Nov 2023).
Strongest evidence
Approved drug backed by large phase 3 trials
Overview

Tirzepatide is a once-weekly peptide that activates GIP and GLP-1 receptors. It is marketed by Eli Lilly as Mounjaro and Zepbound.

What researchers study it for
  • Type 2 diabetes
  • Obesity and overweight
  • Weight-related conditions studied in later trials
Mechanism

Dual agonist of the GIP and GLP-1 receptors - incretin hormones that act on insulin secretion, glucagon, gastric emptying and appetite.

Evidence (2)
  1. Human · randomized trial

    SURMOUNT-1 (phase 3, NEJM 2022)

2,539 adults with obesity or overweight without diabetes, 72 weeks. Mean weight change was −15.0%, −19.5% and −20.9% across the three doses versus −3.1% with placebo. Gastrointestinal adverse events were the most common and mostly mild to moderate.[1]

  2. Approved drug

    FDA approvals

Approved as Mounjaro on May 13, 2022 and as Zepbound on Nov 8, 2023 (Drugs@FDA).[2][3]

Limitations of the evidence
  • Trial results describe the approved medicine in defined patient populations.
  • Products on this site are sold for laboratory research only. They are not the medicines used in the trials above, and those results do not apply to research material.
Questions researchers ask
Is tirzepatide the same as retatrutide?
No. Tirzepatide activates two receptors (GIP, GLP-1); retatrutide also activates the glucagon receptor and is still investigational.
